Kohara clones

Kohara clones are a set of Escherichia coli genomic phage clone library constructed using a lambda phage vector (Kohara et al., 1987). Each clone contains a chromosomal DNA fragment partially digested with Sau3A or EcoRI and cloned in the lambda 2001 vector (for EcoRI library) or the EMBL4 vector (for Sau3A library). Each Kohara clone was digested with 8 restriction enzymes, namely BamHI, HindIII, EcoRI, EcoRV, BglI, KpnI, PstI and PvuII, electorphoresed and aligned according to the positions of the restriction enzyme cutting sites. Consequently, a total of 476 aligned clones were selected to cover essentially the whole of the E. coli chromosome with minimal overlaps that was named as the "minimal set".
